RNA Genome Profiles of Some Sudanese and South African Serotypes of Bluetongue Virus Using Agarose Gel Electrophoresis and SDS-PAGE

Abstract: The double stranded RNA (dsRNA) genome segments of the Sudanese serotypes of Bluetongue virus (BTV) serogroup were analysed by agarose gel and sodium dodecyl sulphate (SDS)- polyacrylamide gel electrophoresis (SDS-PAGE). The dsRNA genome segments profiles of Sudanese BTV serotypes 1, 2, 4 and 16 were compared with those of South African BTV serotypes 1, 19 and 24. Both systems (agarose gel and SDS-PAGE) showed 10 segments, which is characteristic pattern of BTV serogroup. The agarose gel showed identical genome profiles for all Sudanese and South African serotypes of BTV serogroup. However, SDS-PAGE system was able to detect genetic variation between Sudanese and South African BTV serotypes and among the Sudanese serotypes of BTV. The result of this study suggested that, the agarose gel electrophoresis could be used as supportive or complementary method to facilitate tentative diagnosis of BTV infection in susceptible animal populations. In addition, the SDS-PAGE could also be used to detect genetic variations between serotypes of BTV serogroup from different continents.
